Apoptosis 란?

 

– 다세포 생물체에서 일어나는 Programmed cell death의 일종으로 세포 형태와 내부의 생화학적 변화로 인해 세포가 죽는 현상

– 세포의 팽창과 균열, 세포막의 변화, 핵 단편화, 염색질 응축, 염색체 절단, phagocytosis로 종결

– Necrosis와는 다른 cell death로 생물체에 해를 끼치지 않고 발달 및 노화 과정에서 세포 집단을 유지하기 위한 항상성 mechanism으로 생명 주기에 유익

– Signaling pathway에 따라 내인적 세포자살과 외인적 세포자살로 나뉨

Pro-caspase 8(DED Domain), Adaptor FADD (DED Domain and Death Domain), Death Receptor (Death Domain)

 

(1) 내인적 세포자살 (Intrinsic Apoptosis)

– Apoptotic cell은 소멸되기 전까지 세포막의 기능과 생체 기능을 유지하고 있기 때문에 거식세포 또는 다른 세포들의 phagocytic activity 에 의한 빠른 제거 가능

1) 유발 원인: 성장인자 감소, DNA 손상, ER Stress, ROS 과부하, DNA 복제 Stress, Microtubular Alterationᆢ

2) Mechanism : 세포 내부의 손상 → Mitochondria의 Cytochrome C release → Caspase Activation → DNA 절단

 

(2) 외인적 세포자살 (Extrinsic Apoptosis)

– 두 종류의  plasma membrane receptor에 의해 개시

· Death Receptor

· Dependence Receptor (FAS, TNFRᆢ)

– 유발 원인: 세포 외 미세 환경의 변화

2) Mechanism : 세포 외부의 자극이 Death Receptor를 통해 신호 전달 → Caspase Activation → DNA 절단
