Apoptosis(세포사멸) 분석: 주요 기법과 관련 제품 알아보기

Apoptosis란?

  • Apoptosis는 다세포 생물체에서 일어나는 Programmed Cell Death(세포 예정사)의 일종입니다.
  • 세포 형태 변화와 내부의 생화학적 반응을 통해 세포가 스스로 사멸하는 과정을 의미합니다.
  • 핵 단편화, 염색질 응축, 염색체 절단 후 phagocytosis로 정리됩니다.
  • Necrosis와 달리 생물체에 해를 끼치지 않고, 발달·노화 과정에서 세포 항상성 유지에 중요한 역할을 합니다.
  • 세포사멸 경로는 신호 체계에 따라 내인적(Intrinsic)과 외인적(Extrinsic) 경로로 나뉩니다.

(1) 내인적 세포자살 (Intrinsic Apoptosis)

  • Apoptotic cell은 소멸 전까지 세포막과 생체 기능을 유지하므로, 거식세포의 phagocytic activity에 의해 빠르게 제거됩니다.
  • 유발 원인: 성장인자 감소, DNA 손상, ER Stress, ROS 과부하, DNA 복제 Stress, Microtubular Alteration 등
  • Mechanism: 세포 내부 손상 → Mitochondria의 Cytochrome C 방출 → Caspase 활성화 → DNA 절단

(2) 외인적 세포자살 (Extrinsic Apoptosis)

  • 두 종류의 plasma membrane receptor에 의해 개시됩니다.
    • Death Receptor
    • Dependence Receptor (FAS, TNFR 등)
  • 유발 원인: 세포 외부의 미세환경 변화
  • Mechanism: 외부 자극이 Death Receptor를 통해 전달 → Caspase 활성화 → DNA 절단
